该剧主要讲述的是主人公Rick是亚特兰大城郊一座小镇的副警长。在执行公务的过程中，Rick遭到枪击，伤势严重，被人紧急送往当地医院进行抢救。当Rick醒来之后，发现医院里已经空无一人。他意识到外面一定出了大事，但又不清楚到底是什么事。全剧围绕主人公Rick与他的同伴在丧尸蔓延、危机四伏的美国四处逃亡 时的经历展开，剧情扣人心弦。在《行尸走肉》(The Walking Dead)第五季将会确定3个常驻角色，分别是终点站神秘老大Gareth安德鲁·韦斯特、Tara 阿兰娜·莫特森、Rosita克里斯蒂·瑟拉图斯，这也就意味着他们三人在第五季生命在一定程度上是有保障的。Gareth将是第五季主要反派角色，Tara在格伦故事线中作用越发重要，而Rosita则是原著漫画经典角色。另外，制片人还透露，第五季会讲述终点站的历史。
